The American sprint star clocked a stunning 14.67 seconds at the World Athletics Continental Tour Gold event, obliterating the previous world best of 14.92 that had been set earlier this year by Jamaican Olympic silver medalist Kishane Thompson.
Competing over the rarely run distance for the first time in his career, Lyles showcased the speed and endurance that have made him one of the world's most dominant sprinters.
The reigning Olympic 100m champion and four-time world 200m gold medalist were locked in a fierce battle with South Africa’s Sinesipho Dambile coming off the bend before finding another gear in the closing stages to pull away for a commanding victory.
Dambile also dipped under the previous world-best mark, finishing second in 14.78, while Australian teenage sensation Gout Gout completed the podium places in 14.96.
The result marked a notable improvement for the 18-year-old Australian, who endured a disappointing outing at the Oslo Diamond League just a week earlier.
Gout finished sixth in the men's 200m in Norway after a sluggish start from the blocks, clocking 20.60 seconds.
His performance in Ostrava offered signs of a return to form following a breakthrough season highlighted by his remarkable 19.67-second run at the Australian Athletics Championships in April.
That time shattered his own national record and eclipsed the fastest 200m ever recorded by Usain Bolt as a teenager.
For Lyles, the victory further extends his unbeaten run in the 2026 outdoor campaign. The American has now added another landmark performance to a season that has already seen him triumph at the World Athletics Continental Tour Gold meeting in Tokyo and the Wanda Diamond League in Rome.
Elsewhere in Ostrava, Switzerland's Audrey Werro produced another world-class display in the women's 800m. Just nine days after running a world-leading time in Stockholm, Werro dominated the field to win in 1:54.45.
The performance ranks as the eighth-fastest women's 800m time in history and leaves her just 0.48 seconds shy of the Swiss national record she set earlier this month.
Dutch star Femke Bol impressed in her outdoor 800m debut, finishing second in 1:57.13. The time moved the two-time world 400m hurdles champion to third on the Netherlands' all-time list behind only Ellen van Langen and Sifan Hassan.
Another meeting record fell in the men's mile as American Nathan Green produced a strong finishing kick to break a mark that had stood since 2009.
Green surged past compatriot Vincent Ciattei in the closing stages to claim victory in 3:49.44, capping a memorable evening of athletics in Ostrava.
