The Slum Boys have played 16 matches this year, winning only a quarter of the fixtures, with Kamau's final straw being the 2-0 defeat to relegation-threatened APS Bomet on Sunday.
In a brief update, Mathare revealed that assistant coach Mohammed Mchulla has been entrusted with the reins until the end of the season.
"The Club has reached a decision that Coach John Kamau steps aside from his role as Head Coach with immediate effect.
Assistant Coach Mohammed Mchulla will take over as Interim Head Coach for the remainder of the 2025/2026 season," read the statement.
Kamau’s departure brings to an end his second stint with the club, which he joined in August 2024 after leaving Posta Rangers in April of the same year.
The former Thika United coach rejoined the 2008 league champions after their promotion back to the FKF Premier League, taking over from veteran Francis Kimanzi, who had led the team to promotion from the National Super League (NSL).
In the APS match that would eventually cost Kamau his job, the Law Enforcers took the lead in the 28th minute through captain Philip Wasai before Rodgers Kipkemoi added a second in the 75th minute.
However, more concerning for Mathare United fans in the stadium was the team's poor display, with APS wasting numerous chances to win by an even bigger margin.
Prior to that, Mathare were held to a 2-2 draw by relegation-bound Sofapaka, a match in which they threw away a 2-1 lead and conceded in the 96th minute to share the spoils.
Mathare are currently 14th on the log with 35 points, three points more than Ulinzi Stars, who occupy the final relegation slot, albeit with a game in hand.
Next up for the Slum Boys is an away clash against Mara Sugar FC, where Mchulla will be tasked with leading the team back to winning ways.
